I believe its either the fuel pump or the oil pump priming the system. I have the 2.0L Turbo 4cyl and mine makes the same noise upon unlocking the door and I haven't given it a second thought. My wifes v6 Traverse doesn't make the same noise, that I'm aware of, upon unlocking like that.

unlike the others I think something is going on within the door! Has it been making this sound since you picked it up? I have a 20 2LT Blazer myself and have owned several other GM vehicle with the same lock and handle assemblies and have never heard what your hearing. I don‘t believe it is a fuel pump I think it is a mechanical movement within the door. Good luck with your investigation and please post the outcome.

Yeah, it's been making the sound since Day 1. No variations in pitch, tone, or duration. So I think it is a normal operation sound.
I may poke around the door and undercarriage a bit, to see if I can narrow in on the source.
